Summary of methods for lipid analysis. (A) Traditionally, serum cholesterol and triacylglycerol levels have been measured. (B) High performance liquid chromatographic separation using normal-phase techniques and evaporative light-scattering detection enables the separation of major lipid classes. (C and D) Isolation of phospholipid fraction (PL) is performed on silica thin-layer chromatography (TLC) and the fatty acid composition of total serum or its PL fraction is determined by capillary gas chromatography (GC). (E) Two-dimensional GC coupled to time-of-flight mass spectrometry (GC×GC-TOF) is used for small molecule metabolomics and (F–G) liquid chromatography-mass spectrometry (LC-MS) is commonly used in lipidomics. (H) Data mining and processing methods are needed to analyse the data.
