  • Supplementary Figure 1 - (A) Comparison of levels of luciferase activity induced by the endogenous AR-Q865H variant (blue curve) and the exogenous transfected AR-Q865H variant (red curve) in MDA-MB-453 cells treated with increasing concentrations of DHT (0-100 nM). (B) Same data as presented in (A), but with the Y axis scale expanded to show DHT-induced transactivation activity of the endogenous AR-Q865H variant. Data points represent the average ± SEM of at least two independent experiments, each performed at least in quadruplicate.
  • Supplementary Figure 2 - AR, PR and GR levels in MDA-MB-453 cells. (A) MDA-MB-453 and T-47D breast cancer cells were maintained in regular growth media containing 10% fetal bovine serum. Lysates were analyzed for AR, PR and GR levels by Western blotting. (B) MDA-MB-453 cells were plated in phenol red free media containing 10% charcoal stripped FBS and treated 48 hours later with vehicle (veh, 0.1% ethanol, control), DHT (1 nM) or MPA (100 nM). AR (left panel) and GR (right panel) protein were detected by Western blotting. MDA-MB-453 cells growing in media containing phenol red and 10% full serum were used as a positive control. Tubulin was used as a loading control.
